Kinetics of enzyme reactions

-
The aim of the laboratory class:
Measure and evaluate the kinetic parameters of the reaction catalysed by lactate dehydrogenase (LD)
From the measured data determine the maximum reaction velocity (Vmax) and the Michaelis constant (Km).
From the measured data determine the activity of lactate dehydrogenase in the serum sample
Laboratory class – examination topics:
- enzyme structure
- kinetics of enzyme reactions, characterisation of the Michaelis constant ( 1st and zero order reactions), the Lineweaver-Burk plot
- how to determine rate of an enzyme reaction (decrease in substrate, increase in product)
- lactate dehydrogenase
